    摘 要:目的 建立超高效液相色谱-四级杆-飞行时间质谱法快速定性定量检测曼陀罗食物中毒事件中的有毒物质。方法 样品经甲醇提取后,待测液经ACQUITY UPLC? BEH C18色谱柱(2.1mm×50mm,1.7μm)分离,以A相0.01%甲酸水和B相乙腈为流动相,进样量2.0μL,柱温35℃,流速0.2μL/min,经ESI正离子模式采集,以质谱精确分子量和色谱峰保留时间做定性分析,[M]+H为定量离子做定量分析。结果 在1.0~200.0 ng/mL范围内3种有毒物质线性关系良好r2均≥0.990;检出限为1.0~2.24 ng/mL,定量限为0.27~7.46 ng/mL;三水平加标回收率在78%~89%范围内,相对标准偏差(RSD)均≤3.0 %。两起中毒事件中曼陀罗样品检出东莨菪碱含量0.65mg/kg、山莨菪碱0.66mg/kg和莨菪碱0.72mg/kg;曼陀罗泡酒样品中检出东莨菪碱含量0.0048mg/kg、山莨菪碱0.041mg/kg和莨菪碱0.16mg/kg;在数名中毒患者的血液和尿液样本中不同程度检出东莨菪碱、莨菪碱和山莨菪碱。结论 实验室建立的方法快速、高效、准确度高,适用于曼陀罗中毒事件中有毒物质的定性定量检测,两起中毒事件是因食用曼陀罗和曼陀罗泡酒引起的。

    Abstract:

    Abstract: Objective To establish an ultra-high performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC/Q-TOF-MS)method for rapid screening of toxic substances in Datura stramonium L. Methods After the samples were extracted by methanol, the liquid to be tested was separated on ACQUITY UPLC? BEH C18 column (2.1mm×50mm, 1.7μm) with 0.01% formic acid water as mobile phase A and acetonitrile as mobile phase B. The sample volume was 2.0μL, the column temperature was 35℃, and the flow rate was 0.2μL/min. ESI positive ion mode collection, mass spectrometry with precise molecular weight and chromatographic peak retention time for qualitative analysis, [M]+H as quantitative ion for quantitative analysis. Results The linear ranges were 1.0-200.0μg/L with r 2 ≥0. 990. The limits of detection were 0.08-2.24 ng/L the limits of quantification were 0.27-7.46 ng/mL. The recoveries were in the range of 78%~89% and RSDs were ≤3.0%.The contents of scopolamine 0.65mg/kg, anisodamine 0.66mg/kg and hyoscyamine 0.72mg/kg were detected in Datura stramonium L collected in the field. Scopolamine 0.0048mg/kg, anisodamine 0.041mg/kg and hyoscyamine 0.16mg/kg were detected in the liquor of Datura stramonium L. The target compounds were detected in different degrees in the blood and urine of the poisoned patients. Conclusion The method established in the laboratory is fast, efficient and accurate, which is suitable for the qualitative and quantitative detection of toxic substances in the poisoning incidents of Datura stramonium L. Two poisonings were caused by consumption of Datura stramonium L.
