    • Preliminary assessment of recombinant human lactoferrin for allergenic potential

      Abstract:To evaluate the allergenic potential of recombinant human lactoferrin (rhLf) expressed by genetically modified cow in milk.Methods Bioinformatics searches and digestibility assay were conducted to investigate the allergenic potential of rhLf. [WT5"HZ]Results Bioinformatics searches showed that there was high-level similarity between rhLf and some known allergens, such as lactotransferrin and ovotransferrin in amino acid sequence, and Asp f 2, Ole e 10and Ole e 9in structure. Digestibility assay showed that rhLf broke down into small fragment rapidly during gastric fluid digestion, while it was resistant to intestinal fluid digestion.ConclusionRhLf has allergenic potential.

    • The effect of exposure to transgenic Bt rice on reproductive system of male offspring rats

      Abstract:To investigate the effect of transgenic Bt rice (TT51) exposure on reproductive system development of male filial generation of Wistar rats. MethodsMarket rice, TT51and its parent were continuously administered to the female and male rats of parental generation for 70days, and then produced the filial generation. The female parents were continued to feed with test samples during gestation and lactation. Filial generation male rats were administered with ordinary feed for 70days after weaned, and body weight, food consumption and growth status were recorded every week. The young male rats were sacrificed on the 70th day for blood routine, blood biochemistry, serumal hormone level, reproductive organ weight , sperm parameter and other target examinations. [WT5"HZ]Results No significant defference was observed between the TT51rice , market rice and its parent rice rice groups on body weight, food consumption, blood routine, blood biochemistry, serumal hormone level, sperm parameter. ConclusionTT51rice exposure had no observed adverse effect on the reproductive system development of male filial generation.

    • Correlation analysis on migration of two polymer monomers in melamine tableware

      Abstract:To investigate the migration and correlation of melamine and formaldehyde from melamine tableware in China markets, and to provide the scientific basis for future risk assessment. Methods Migration of formaldehyde was analyzed by spectrophotometric and migration of melamine was analyzed by HPLC. Migration experimental conditions were 90degree for 30minutes, and the analog solvent was 3% acetic acid. [WT5"HZ]Results 41samples were collected from markets. Melamine was detected in 28samples and the maximum migration value was 0.85mg/dm2. It was about 17.0% of SML regulated by EU. Formaldehyde was detected in all 41samples. The maximum migration value was 270.56mg/dm2. It was about 108.2times SML regulated by EU. 53.66% of samples exceeded the SML value. Conclusion If melamine wasn’t detected but high amount of formaldehyde was detected, then the tableware might not be melamine tableware. The safety of melamine tableware is worthy of further attention.

    • Effect of diisobutyl phthalate on active avoidance behavior and oxidative stress of brain tissue in mice

      Abstract:To give a preliminary research on diisobutyl phthalate(DiBP) on active avoidance behavior, and the potential of free radical oxidation damage in brain tissues of mice. Methods Accommodating 60Kunming mice to the animal room for 3days, then dividing the mice into 5groups according to their weights including one control group and four experimental groups were 50,0, 500and 1000mg/kg BW. The mice were fed with corn oil in control group, and the other groups were fed with different dose of diisobutyl phthalate mixture by gavages last for 8weeks. At the end point, active avoidance response was examined, then all mice were sacrificed and biomarkers in the brains, such as SOD, GSH-Px and 8-OHdG were testeds.[WT5"HZ]Results In the active avoidance response test, the mice of groups 500and 1000mg/kg BW showed longer response latency than control group(P<0.05) and the mice of group 1000mg/kg BW showed fewer conditioned response than control group(P<0.01); the mice of groups 500and 1000mg/kg BW showed a lower conditioned response rate than control group(P<0.05); the GSH-Px activities in groups 250and 1000mg/kg BW significantly decreased (P<0.05), and the 8-OHdG content increase significantly comparing to the control group, (P<0.05). [WT5"HZ]Conclusion Exposure to DiBP could decrease active avoidance ability. Oxidative stress could be induced by DiBP in mice brain tissue.

    • Establishment of the typing method by fluorescent amplification fragment length polymorphism for Pseudomonas cocovenenans subsp. farinofermentans

      Abstract:To establish a genotyping method for Pseudomonas cocovenenans subsp. farinofermentans with fluorescent amplification fragment length polymorphism(FAFLP).Methods Four low-frequency-cleavage restriction endonucleases (ApaⅠ, EcoRⅠ, Hind III, PstⅠ) and two high-frequency-cleavage restriction endonucleases(MseI, TaqⅠ)were tested to optimize the combination for FAFLP. Pre-selective and selective PCR reactions were then optimized. The selective PCR products was separated on LI-COR 4300DNA Analyzer and digital imagines were analyzed by BioNumerics software. In addition, the FAFLP method was compared with other two typing methods(VITEK 2and 16s rDNA).[WT5"HZ]Results 19isolates were clustered into 7groups with 80.85% as the minimum similarity and 98.77% the maximum among groups using the combination of ApaⅠ-G/TaqⅠ-G. The isolates were clustered into 10groups with 48.68% as the minimum similarity and 99.67% the maximum among groups using the combination of PstⅠ-T/TaqⅠ-G. The two combinations of FAFLP could descriminate all the strains in the study, but not the other two methods(VITEK 2and 16s rDNA).ConclusionThe two combinations of FAFLP both had a good discrimination and could be applied for tracing Pseudomonas cocovenenans subsp. farinofermentans in the investigation of foodborne diseases.

    • Study on the dietary protein requirements of young female adults

      Abstract:Reference intakes of nutrients are the basis of nutritional research. The results of The Nutrition and Health Status Survey of Chinese Residents in 2002discovered that dietary reference intakes of protein in healthy adults were overestimated and need to be reevaluated. Methods 9healthy female adults received a restricted daily level of protein intake 0.75,0.82,0.89,0.97and 1.05g/(kg·d). Every dietary protein level lasted six days. On the sixth day, they took 13C-leucine as a tracer to carry out sTable isotope metabolic experiment. The kinetic parameters including leucine flow, oxidation rate and oxidate production rate were calculated. We applied biphasic linear regression crossover analysis on 13CO2 data and protein intakes. We identified a breakpoint at the minimal rate of appearance of 13CO2 expiration, specific to each level of dietary protein intake. [WT5"HZ]Results The Estimated Average Requirement (EAR) and the Recommended Nutrient Intake (RNI) of protein for healthy Chinese young female adults were determined to be 0.85and 0.97g/(kg·d). Conclusion According to the reference weight of female adults in recent national nutrition survey, we calculated that the daily protein recommended requirements for female adults was 55g, which was lower than current protein recommended requirements.

    • Effects of strontium mineral water on proliferation and function of human umbilical vein endothelial cells

      Abstract:To observe the effects of strontium mineral water (SMW) on proliferation and secretion of human umbilical vein endothelial cells (HUVEC) at different strontium concentration. [WT5"HZ]Methods HUVEC were cultured in vitro. After culturing in a normal Dulbecco''s Modified Eagle''s Medium (DMEM) for 18hours, the cells were transferred to the conditioned medium with 2,4,6,8mg/L strontium mineral waters and double distilled water (DDW) treatment. The proliferation of cells was observed by Cell Counting Kit-8(CCK-8) assay at 24th, 48th, 72nd and 96th hours. Cell growth was observed by hematoxylin staining at 72nd hours. Meanwhile, cell protein was measured by Enhanced BCA Protein Assay kit and levels of extracellular vein endothelial growth factor (VEGF), nitric oxide (NO) and endothelin (ET) were tested by enzyme-linked immuno sorbent assay (Elisa). [WT5"HZ]Results The proliferation activity of cells in 2and 4mg/L SMW groups were significantly increased (P<0.05), while significantly decreased in 8mg/L SMW group (P<0.05) after 72hours. Total of extracellular VEGF in SMW groups were lower than DDW group (P<0.05), and there was no significant difference between SMW groups (P>0.05) after 72hours. NO and ET of cells in 2and 4mg/L groups were hyposecretion, while 6and 8mg/L groups were on the contrary. NO/ET ratio of 4mg/L SMW group was higher than the others (P<0.05), and the difference of NO/ET ratio between the other groups was not significant (P>0.05). [WT5"HZ]Conclusion A certain concentration of strontium in mineral water is good for proliferation of HUVEC. It could affect HUVEC secretion function of ET and NO, and decrease vascular tone. VEGF was not the direct factor which enhanced the proliferation activity of HUVEC culturing in strontium mineral water.

      Abstract:To evaluate the influence of sFat-1transgenic pork on the blood routine and biochemical and immune indicators of mice.Methods Granular compound feed with low and high dosage of sFat-1transgenic pork or pork control and basal feed control were administered orally to five groups of mice respectively for 30days. Blood routine and biochemical indicators, immunoglobulin and the percentage of lymphocytes in peripheral blood were observed.Results There was no significant difference between the five groups on blood routine and immunoglobulin; the serum BUN concentration was significantly higher in high-dosage sFat-1transgenic pork group than control group in both male and female (P<0.05); the percentage of lymphocytes (CD3+CD4+) in peripheral blood of female was significantly higher in low-dosage sFat-1transgenic pork group than low-dosage pork control group (P<0.05), while the percentage of lymphocytes (CD3+CD8+) of female mice was significantly higher in high-dosage sFat-1transgenic pork group than low-dosage pork control group(P<0.05).Conclusion There was no influence of sFat-1transgenic pork on blood routine and biochenicnt and immune indicators of mice at the given dosage in the study.

    • Analysis of serologic traceability results in two cases of food poisoning caused by Vibrio parahaemolyticus

      Abstract:To trace the serological source of two food poisoning cases caused by Vibrio parahaemolyticus(VP) and analyze the relationship between serotypes in the suspect food and patient samples.Methods Based on the GB/T 4789.7-2008method, the detected VP was serotyped, and hemolysin tests were performed. The directly heat-resistant hemolysin gene(tdh), tdh-related hemolysin gene(trh) and toxins regulated genes(toxR) were amplified by PCR. Results9VP serotypes in two food poisoning cases were identified by expanding the identification of suspicious colonies in the sample. There were 13strains of O3∶K6,6strains of O2∶K28,2strains of O1∶K56. The results of tdh gene detection were positive in 17strains from 27isolated VP strains which were consistent with the hemolysin tests.Conclusion Expanding the identification of suspicious colonies contributed to the traceability of food poisoning caused by VP. Although O3∶K6was the major pathogen causing food poisoning, VP serotypes from different samples show diversity. The tdh gene detection is equivalent to the hemolysis test in detecting the VP pathogenicity.

    • Simultaneous determination of five aluminum lake dyes in coated candy by ultra performance liquid chromatography

      Abstract:To develop a method for simultaneous determination of tartrazine Al lake, sunset yellow Al lake, ponceau 4R Al lake, allura red Al lake and brilliant blue Al lake in coated candy by ultra performance liquid chromatography. MethodsTarget analytes in coated candy were extracted by sodium hydroxide solution, separated by gradient elution with anetonitrile and 10mmol/L acetic ammonium as mobile phase, and detected by photodiode array detector. ResultsGood linearities were achieved for five aluminum lake dyes in the range of 0.25-0.50mg/L, with correlation coefficients R2>0.999. The quantification limits of the method were 1.0mg/kg. Mean recoveries of five target analytes (spiked at three concentration levels) ranged from 79.5% to 116.4%, with relative standard deviations between 0.92% and 6.48%. Conclusion This simple, rapid and accurate method could be applied in simultaneous detection of five aluminum lake dyes in coated candy.

    • Determination of benzo(a)pyrene and its metabolite in seafood by high performance liquid chromatography with fluorescence detection

      Abstract:An analytical method for the determination of benzo(a)pyrene and its metabolite 3-hydroxybenzo(a)pyrene in seafood was developed by high performance liquid chromatography with fluorescence detection (HPLC-FD).Methods The samples were extracted by 65% acetonitrile, treated by vortex blending and centrifugal precipitation. For benzo(a)pyrene and 3-hydroxybenzo(a)pyrene, the chromatographic separation were achieved in CenturySIL C18-BDS(150mm×4.6mm, 5μm)column with methanol-water(pH 4.5)(85∶15),(87∶13)as mobile phase with a flow rate of 1.0ml/min, respectively. The benzo(a)pyrene and 3-hydroxybenzo(a)pyrene were detected under FD at λexem=265nm/450nm, λexem=365nm /450nm with an injection volume of 20μl, respectively.Results Benzo(a)pyrene was linear in range of 0.1~10.0ng/ml,r=0.9994and 3-hydroxybenzo(a)pyrene was linear in range of 1.07~100.7ng/ml,r=0.9921. For benzo(a)pyrene and 3-hydroxybenzo(a)pyrene, the limit of quantification was 0.1μg/kg, 0.2μg/kg, the recoveries were 96.8%~112.0%, 72.6%~83.3%, and the relative standard deviations was 6.0%, 5.2%(n=5), respectively.Conclusion The method is simple,rapid and highly sensitive. It is suitable for the determination of benzo(a)pyrene and 3-hydroxybenzo(a)pyrene in seafood.

      Abstract:To establish a rapid method for simultaneous determination of 7preservatives (sorbic acid, benzoic acid, dehydroacetic acid, methyl p-hydroxybenzoate, ethyl p-hydroxybenzoate, propyl p-hydroxybenzoate, butyl p-hydroxybenzoate) and 2sweeteners (saccharin sodium, acesulfame potassium ) in pickled vegetables by ultra performance liquid chromatography.MethodsWaters ACQUITY UPLC BEH C18 (2.1mm×100mm, 1.7μm) was used as separation column, and methanol and 0.02mol/L ammonium acetate was used as mobile phase in gradient elution mode at the flowrate of 0.20ml/min. The column temperature was set to 35℃ and detected by diode array detector at 230and 257nm, respectively. ResultsThe 9food additives were completely separated within 15mins. In the range of 10~250mg/L, the peak area and content was in a good linear relationship (r≥0.9991). The average recoveries at three spiked levels were 85.1%~98.8%, with RSDs of 3.5%~8.5% (n=6). The limits of detection (S/N=3) were 0.2~1.0mg/kg and the limits of quantification (S/N=10) were 0.5~4.0 mg/kg.Conclusion This method is simple and fast with high sensitivity, and it is suitable for the determination of food additives in pickled vegetables.

      Abstract:A method of high performance liquid chromatography was established for simultaneous determination of salidroside and adenosine in healh food.Methods Salidroside and adenosine in samples were extracted ultrasonically with 50%methanol as the solvent and separated on a shim-pack CLS-ODS column(150mm×4.6mm,5μm);the mobile phase consisted of methanol+20mmol/L Potassium dihydrogen phosphate(15+85,V/V). The flow rate was 1.0ml/min; the injection volume was 10μl; the UV detection wavelength of salidroside and adenosine ware 275and 260nm. Results The standard curve of salidroside was linear over the range in 10.0~200μg/ml(r=0.9995).The recovery was 92.5%~99.5%,RSD 2.8%~4.7% (n=6);The standard curve of adenosine was linear over the range in 2.50~50.0μg/ml(r=0.9995).The recovery was 90.0%~94.0%,RSD 2.1%~5.2%(n=6). The detection limits of salidroside and adenosine were 0.01and 0.005μg/g.Conclusion The method was simple, sensitive and accurate, it could be used for simultaneous determination of salidroside and adenosine in health food.

    • Determination of melatonin in Cordyceps sinensis mycelium by high liqiud chromatography electrospray inonization tendem mass spectrometry

      Abstract:To establish a method of determining melatonin in Cordyceps sinensis mycelium by high performance liquid chromatograhy electrospray inonization tendem mass spectrometry(HPLC-MS/MS).Methods Melatonin in the sample was extracted utrasonically with methanol. The content was then seperated on Thermo Beta Basic-18column (150mm×4.6mm, 5μm) with methanol/ 0.1% formic acid solution (60∶40) as mobile phase. The flow rate was 0.6ml/min, and column temperature was set to 40℃. Detection was performed under positive ion electrospray ionization tandem ion trap mass spectrometry (ESI+)-tandem mass spectrometry (MS/MS) mode.Results There was a good linearity of melatonin in the range of 10~200μg/kg with correlation coefficient of 0.9992. The detection limit of the method was 0.5μg/kg, and the limit of the quantification was 2.0μg/kg. The averange recovery was 81.92%. The relative standard deviration was lower than 10%.Conclusion This method is selective and fast. It can be applied to detect the melatonin in Cordyceps sinensis mycelium powder, and it may also provide basis for the determination in other complex systems.

Methods Develop a questionnaire investigation and use SPSS software for statistic analysis. ResultsThe FL TBT of America, EU and Japan are more strict than the others, GMO label, allergen label and nutrition label are main TBT requirements; FL TBT brings a lot of difficulties to our export industries; they have various needs for countermeasures.Conclusion We should attach importance to FL TBT and respond proactively.

    • Validation of proficiency test of aflatoxin B1 in peanut oil

      Abstract:This assessment program was established by Guangdong Inspection and Quarantine Technology Center (IQTC) to evaluate the detection capability of aflatoxin B1 in peanut oil approved by Certification and Accreditation Administration of the People''s Republic of China (CNCA). MethodsThe assessment was performed according to ISO 13528:2005, CNAS-GL02:2006, CNAS-GL03:2006 and the international harmonized protocol for proficiency testing. The homogeneity of samples was assessed by F-test and S S≤0.3σp,the satiability was assessed by t-test. The median was used as assigned value to calculate a z-score which was used as a numerical indicator to show participants’ performance in the program.ResultsLaboratories which were identified as “unsatisfactory or questionable” had one chance for a second-round test. Finally, 88samples were observed as satisfactory, 1sample as questionable and 2samples as unsatisfactory. Conclusion Most of domestic related laboratory possess good detection capability of aflatoxin and could meet the requirements of international proficiency testing standards.

    • Ananlysis of Pb, Hg and As in health foods in Henan province from 2008to 2011

      Abstract:To understand the contamination status of Pb, Hg and As in health foods. MethodsHealth food samples from 2008to 2011were tested for Pb, As and Hg by national standard methods, and data were analyzed with SPSS 13.0software. ResultsThe detection rates of Pb, As and Hg among 851samples were 32.37%, 66.67% and 63.83%, respectively. 8.42%, 7.97% and 1.86% of the samples failed to pass the national standars, with means of 1.03,0.44and 0.08mg/kg for Pb, As and Hg, respectively. The descending order for Pb and Hg contamination was plant products, animal products and nutritional supplements. The descending order for As was animal products, plant products and nutritional supplements. The descending order for Pb in different formulation was pills, granules, capsules, tablets and liquids, while for As and Hg was pills, capsules, granules, tablets and liquids. 59.93% of the samples were positive for more than one contaminant of Pb, Hg and As,Pb and Hg were detcted simultaneously in 44.90% of the samples. ConclusionContamination of Pb, Hg and As in health foods were common and serious, and mainly related to food materials fro, plants and animals.

      Abstract:To find out the aluminum content of some common flour foods in Nanning, search the influencing factors and provide basis for the regulatory authorities to develop control measures. Methods 28crullers, 32cakes and 10pieces of puffed food were randomly collected from the retail stores of hotels, schools, and booths and then tested and evaluated. ResultsThe aluminum content of three common flour foods was in the range of 42.8-1206mg/kg, and the average was 336.7mg/kg. The aluminum content of 90% cruller was in the range of 320-1206mg/kg. The overall over standard rate of the three flour foods was 60%, and was 92.9%, 46.9% and 10.0% for cruller, cakes and puffed food, respectively. The over standard rate of cruller and cakes from retail stores of hotels, schools and booths was 65.8% (25/38), 53.8% (7/13) and 100.0% (9/9), respectively. The on-site products had a higher aluminum content than the prepackaged puffed food. Conclusion The aluminum content of flour foods in Nanning was seriously over standard, and further actions to strengthen food safety supervision was highly recommended.

      Abstract:To study the epidemiologyical characteristics of botanical food poisoning outbreaks in Shaoguan and provide scientific basis for prevention. Methods Descriptive epidemiological method was applied to analyze the data of botanical food poisoning outbreaks during 2002-2011.Results Thirty-one outbreaks happened with 384sufferers and 8dead cases during 10years. summer was the higher seasons of food poisoning and location of the food poisoning was mainly at home and in school. School students were high-risk population. The main factor of outbreaks were poisonous mushrooms and lagenariasiceraria,but gelsmium elegans and poisonous mushrooms were the leading causes of death.ConclusionIn order to improve the prevention and control effect,it is necessary to strengthen food poisoning prevention in the schools, and enhance the food safety health education.

      Abstract:Multiple-locus variable number tandem repeat analysis (MLVA) is a molecular subtyping technique characterized by the trait of variable number tandem repeat (VNTR). This method is simple and rapid with high-throughput and great capability to distinguish different strains, and has been widely used in molecular typing of a variety of bacteria.       Abstract:Foodborne disease is one of the major global public health concerns. It has also become the largest food safety issue in China. Estimating the frequency of acute gastrointestinal illness in the population is a central issue in evaluating the burden of foodborne diseases. Several countries have conducted studies to estimate the burden of acute gastrointestinal illness and foodborne disease, and China conducted a pilot study designed to estimate the burden of disease during 2010and 2011. In this review, domestic and foreign researches on estimation of the burden of acute gastrointestinal illness and foodborne disease were outlined.
